Polygenic risk scores in epilepsy

چکیده
Abstract An epilepsy diagnosis has large consequences for an individual but is often difficult to make in clinical practice. Novel biomarkers are thus greatly needed. Here, we give overview of how thousands common genetic factors that increase the risk can be summarized as polygenic scores (PRS). We discuss current state research on PRS serve a biomarker epilepsy. The high heritability forms epilepsy, particularly generalized indicates promising potential and prediction. Small sample sizes low ancestral diversity genome-wide association studies show, however, need larger more diverse before could properly implemented clinic.

Multiethnic polygenic risk scores improve risk prediction in diverse populations.
Methods for genetic risk prediction have been widely investigated in recent years. However, most available training data involves European samples, and it is currently unclear how to accurately predict disease risk in other populations. Previous studies have used either training data from European samples in large sample size or training data from the target population in small sample size, but...
متن کاملEffect of polygenic risk scores on depression in childhood trauma.
BACKGROUND Research on gene × environment interaction in major depressive disorder (MDD) has thus far primarily focused on candidate genes, although genetic effects are known to be polygenic. AIMS To test whether the effect of polygenic risk scores on MDD is moderated by childhood trauma. METHOD The study sample consisted of 1645 participants with a DSM-IV diagnosis of MDD and 340 screened ...
متن کاملModeling Linkage Disequilibrium Increases Accuracy of Polygenic Risk Scores.
Polygenic risk scores have shown great promise in predicting complex disease risk and will become more accurate as training sample sizes increase. The standard approach for calculating risk scores involves linkage disequilibrium (LD)-based marker pruning and applying a p value threshold to association statistics, but this discards information and can reduce predictive accuracy. We introduce LDp...
متن کاملPower and Predictive Accuracy of Polygenic Risk Scores
Polygenic scores have recently been used to summarise genetic effects among an ensemble of markers that do not individually achieve significance in a large-scale association study. Markers are selected using an initial training sample and used to construct a score in an independent replication sample by forming the weighted sum of associated alleles within each subject. Association between a tr...
متن کاملPolygenic hazard scores in preclinical Alzheimer disease.
Identifying asymptomatic older individuals at elevated risk for developing Alzheimer disease (AD) is of clinical importance. Among 1,081 asymptomatic older adults, a recently validated polygenic hazard score (PHS) significantly predicted time to AD dementia and steeper longitudinal cognitive decline, even after controlling for APOE ɛ4 carrier status. Older individuals in the highest PHS percent...
